Abstract

Bt Strain S2685-1 was identified by Dr. X. J. Fang’s group from Hainan Institute of Tropical Agricultural Resources (HITAR) in 2009 through 2014. The isolate (isolate No: 20070720S2685) was discovered from soil sample that were collected from Heilongjiang Liangshui Natural Reserves of China. Bt S2685-1 produces dominant 130 kDa crystalline protein with significant larvicidal effects on corn earworm (Helicoverpa zea). Scanning electron microscope observation showed that the strain generates high-yielding bipyramidal crystals during sporulation. SDS-PAGE analysis showed that the protein profile mainly contains six proteins with the molecular weight about 130 kDa, 80 kDa, 75 kDa, 45 kDa, 37 kDa, 25 kDa. The mega-plasmid profile of the Bt S2685-1 showed that it had three mega-plasmid (70 kb, 97 kb, 200 kb). The genome sequencing predicts that it contains cry1A-like gene. Interestingly, the bioassays were carried out on mosquitoes in Diptera and corn earworm (Helicoverpa zea) in Lepidoptera, the results exhibit that the strain Bt S2685-1 is significantly toxic to corn earworm (Helicoverpa zea) larva, but not in mosquitoes. Up to now, we haven’t got the predicted expression products, so, what we need to do next is cloning and expressing the toxic genes, and further focus on other Lepidoptera or Coleoptera insects in agriculture.
